The study of Lewy body disorders will be greatly advanced with the development and characterization of transgenic mouse models. Although most lines overexpress alpha-synuclein, not all lines have necessarily recapitulated LB-like pathology. It is also unfortunate that it has been difficult to model dopaminergic neuron loss such as seen in Parkinson's disease. This review gives a nice overview of what has been done and (hopefully) can stimulate some bright investigator to figure out what we are all doing wrong.
