Findings

This protein-truncating variant was identified in an individual with probable Alzheimer’s disease from the Centre National de Référence - Malades Alzheimer Jeunes (CNR-MAJ), the French national reference center for young Alzheimer patients (Schramm et al., 2022). The carrier’s age of onset was 65 years and their APOE genotype is E3/E4.

One of the proband’s parents also was diagnosed with AD, with age of onset 70 years. The other parent was cognitively intact at age 90, as were two half-siblings—one from each of the proband’s parents—at ages 72 and 80. Genotype information was not available from family members.

In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, including CNR-MAJ, this allele was observed once among the AD cases (Holstege et al., 2022).
