Findings
This rare variant was found in both cases and controls of European ancestry.
In a study that included 15,808 Alzheimer’s cases and 16,097 control subjects from multiple European and American cohorts, this allele was observed 64 times—31 times among the AD cases and 33 times among the controls (Holstege et al., 2022). A mega-analysis of these data did not find an association between the variant and AD risk. This result confirmed the findings of two relatively small studies in which the variant did not associate with AD (Fernández et al., 2016; Sassi et al., 2016).
Functional Consequences
The variant was predicted to be tolerated by SIFT and benign by PolyPhen-2, and it was classified as a polymorphism by Mutation Taster (Sassi et al., 2016).
