Findings

This rare variant has been seen in both Alzheimer's cases and controls subjects of European ancestry and was found to associate with AD in a mega-analysis of nearly 32,000 subjects drawn from multiple European and American datasets (Holstege et al., 2022).

In a pan-European cohort of 1255 Alzheimer’s cases and 1938 controls from the European Early Onset Dementia Consortium, one patient was found to be a heterozygous carrier of this variant (Verheijen et al., 2016). This Swedish carrier was 48 years old at symptom onset, with an unknown family history of AD.

Another AD case was also found to be a heterozygous carrier in a Dutch sample of 640 cases and 1268 controls (Holstege et al., 2017). The cohorts comprising this sample contributed to the 2022 mega-analysis cited above.

The E560K variant was reported in three of 5198 AD cases and two of 4491 controls in a dataset from the Alzheimer’s Disease Sequencing Project (ADSP), consisting of subjects of non-Hispanic Caucasian ancestry from whom whole-exome sequencing data were available (Campion et al., 2019). The ADSP also contributed to the 2022 study.

The variant was classified as likely benign by the criteria of Holstege et al. (Holstege et al., 2017).

Functional Consequences

The variant was predicted to be tolerated by SIFT, neutral by Mutation Taster, and benign by PolyPhen-2 (Campion et al., 2019).
