Findings
This rare variant was found in a genetic screen. It was identified in one control individual of Mandenka origin, a west African ethnic group (Guerreiro et al., 2010). It was absent from the gnomAD variant database (v2.1.1, Oct 2021).

Not applicable.
Biological Effect
Mouse neuroblastoma cells expressing this variant secreted slightly more Aβ42 and similar amounts of Aβ40 compared with cells expressing wild-type APP. The Aβ42/Aβ40 ratio was not significantly altered (Hsu et al., 2020). This variant's PHRED-scaled CADD score, which integrates diverse information in silico, was above 20, suggesting a deleterious effect (CADD v.1.6, Oct 2021).
