Overview

In initial testing on western blots, antibody 6B9 reacted with human ApoE3 and ApoE4 (data not shown) overexpressed in HEK293 cells. As the antibody was generated against an epitope at the N-terminus of human ApoE common to all three isoforms, it is expected to also detect ApoE2. Antibody 6B9 was also shown to stain THP1 cells, a human monocyte cell line derived from a patient with acute monocytic leukemia.

Antibody 6B9 recognizes human ApoE3 over expressed in HEK293 cells. HEK293 cells were transfected with empty vector (-) or C-terminally myc-Flag-tagged ApoE3 (E3). Western blots of supernatants from transfected cells were probed with 6B9 or a commercial antibody from Santa Cruz (SC-E8, #sc-393302). [Courtesy of Christian Haass' laboratory.]

6B9 immunofluorescence in THP1 cells. THP1 cells immunostained with antibody 6B9, followed by a fluorescently-conjugated secondary antibody (red); nuclei counterstained with DAPI (blue). [Courtesy of Trevor Atkeson, Beth Stevens' laboratory.]

Generation and epitope mapping

This antibody was generated in rats immunized with a peptide from the N-terminal region of human ApoE, coupled to ovalbumin. Epitope mapping has not been reported.

Specificity

6B9 recognizes human ApoE.

Validation
