Overview

Monoclonal antibody 6F7 reacts with mouse ABI3. In initial testing, the antibody was shown to work as a detection reagent in western blot and as a capture antibody for immunoprecipitation.

Antibody 6F7 recognizes murine ABI3 overexpressed in HEK293 cells. A) Western blot of lysate of HEK cells transfected with empty vector (mock), human ABI3, or murine Abi3, probed with hybridoma supernatant. 6F7 recognizes murine, but not human, ABI3. B) 6F7 (protein A affinity-purified) immunoprecipitates murine ABI3 expressed in HEK cells. Immunoprecipitation with rat IgG2a served as a negative control. Input was lysate from HEK293 cells transfected with murine Abi3; cells transfected with empty vector (-) served as negative control. Generation and epitope mapping

This antibody was generated in rats immunized with a peptide from the C-terminal region of murine ABI3, coupled to ovalbumin. Epitope mapping has not been reported.

Specificity

On western blots, the antibody recognizes mouse, but not human, ABI3 overexpressed in HEK293 cells.

Validation

6F7 has been tested against Abi3 knockouts in western blots. The antibody detected a band in lysates of spleens from wild-type mice but did not recognize proteins in spleen lysates from Abi3 knockouts.
