This provides histochemical evidence to support a role of the neprilysin in amyloidogenesis of AD brain. The immunoreactive intensity was not strong but favors its pathological involvement.

"Neprilysin is a protease that degrades Ab and is emerging as an important regulator of Ab accumulation. The study of neprilysin represents a novel approach towards understanding Alzheimer’s pathophysiology, and understanding its biology represents a growth area for research in AD."

REAGENTS/MATERIAL:
Brains from 15 patients with AD, three with diffuse Lewy body disease (DLBD) with AD pathology, two DLBD without AD pathology, two tangle only dementia, one schizophrenic, and three control subjects. Mouse monoclonal antibody SN5c/L4-1A1 against neprilysin, IgG, (Ancell) Mouse monoclonal antibody MEM78 against neprilysin, IgG, (Southern Biotechnol) Mouse monoclonal antibody FR4D11 against neprilysin, IgG, (NeoMarkers) Rabbit polyclonal antibody R37 raised against syntehtic peptide corresponding to residues 681-695 of APP695, obtained from Dr. Tsuyoshi Ishii (Sagamidai Hospital)

FUTURE DIRECTION:
More sensitive and quantitative methods needed to further investigate the relationship of histochemical localization of neprilysin and Abdeposition in brain, esp. in brain tissues with heavy Ab burden.
