Name: NIC5-15
Therapeutic Applications: mild to moderate Alzheimer's disease
Therapy Types: small molecule
Mechanisms: NIC5-15 is a naturally occurring anti-diabetic agent with insulin-sensitizing properties. It has also been shown to act as a gamma-secretase inhibitor that spares Notch.
Development Status: investigational in U.S.
FDA Phase: Phase II/IIa/IIb
Primary Medical Role: Administration of NIC5-15 for 6 months decreased Aβ40 and Aβ42 levels in the cerebral cortex and hippocampus of insulin resistant Tg2576 mice. Reduced Aβ levels were due to a reduction in γ-secretase activity. Notch-1 levels were unaffected. In addition to improving Aβ levels, NIC5-15 was reported to improve cognitive functioning in Tg2576 mice at 12 months of age (Wang et al., 2005).
