With background in physics and molecular biology spent first 15 years of research in the field of virology. Worked with Bunya- and Arenaviruses, and later with Hantaan viruses. In 1986 discovered and mapped with co-workers the APP gene and since then stayed in the AD field. Lost a bet to John Hardy by discovering the first PrP mutation in CJD (I did not believe that PrP mutations are related to CJD). Showed involvement of proteasome in APP degradation. Correctly predicted the association of ApoE isoforms with amyloid load. Postulated and proved the sequestration hypothesis of Abeta. Made APP movies in real time in live cells. |
