How specific is  4G8 for insoluble amyloid fibrils in amyloid plaques? I'm getting significant intracellular IR (suggesting intracellular amyloid presence) while using 4G8 to stain for extracellular amyloid.

Professor Irina Alafuzoff of Kuopio University Institute of Clinical Medicine, Kuipo, Finland, has very graciously sent us the following information (professor Alafuzoff is a member of the Alzheimer Research Forum):
 
"We have dealt with this problem in a recent MS we sent to Laboratory Investigation. I hope it will be concidered for acceptance as this issue surely needs clarifications. 
 
"Shortly, 4G8 is not specific as it cross reacts with APP and thus even in a child one will see a lot of intracellular labelling. Based on biochemical analyses all or at least most of the intracellular labelled protein is in soluble form. The only way to test this issue in a study is in addition to an amyloid-beta antibody includ antibodies to APP C and N terminals. Regarding the solubility one has to carry out biochemical analyses."
